Overview

OE226-iNK cells are **induced pluripotent stem cell-derived natural killer (iPSC-NK) cells engineered to overexpress CD226**. CD226 is an activating receptor on NK cells that recognizes ligands such as CD112 and CD155 on tumor cells, leading to NK cell activation and cytotoxicity. Acute myeloid leukemia (AML) cells can induce CD226 downregulation as an immune escape mechanism. By overexpressing CD226, OE226-iNK cells demonstrate **enhanced cytotoxic function and cytokine release against AML cells**. Preclinical models show that OE226-iNK cells significantly reduce leukemia burden, prolong survival, and diminish systemic inflammation compared to unmodified (control) iPSC-NK cells, indicating their **potential as an off-the-shelf immunotherapy for AML and other CD226 ligand-expressing cancers**[1].
