Overview

This combination regimen consists of **ofatumumab** (a fully human anti-CD20 monoclonal antibody), **cyclophosphamide** (an alkylating agent), **doxorubicin** (an anthracycline antineoplastic), **vincristine** (a vinca alkaloid antineoplastic), and **prednisolone** (a corticosteroid). It is designed as an immunochemotherapy protocol, often termed ofatumumab plus "miniCHOP" when reduced doses are used. This combination is primarily investigated for elderly patients with **diffuse large B-cell lymphoma (DLBCL)** and may be used for other B-cell non-Hodgkin lymphomas. Ofatumumab targets and depletes CD20-expressing B cells. Cyclophosphamide, doxorubicin, and vincristine each disrupt cancer cell proliferation via different cytotoxic mechanisms, while prednisolone induces apoptosis and immune modulation. The regimen has demonstrated improved survival and reduced toxicity in older, frail patients compared to similar regimens using rituximab, and serves as an alternative where rituximab is unsuitable or for clinical trial protocols[1][3][7].
