Overview

Ofatumumab + dacarbazine is a combination regimen consisting of two distinct anticancer agents. Ofatumumab is a fully human monoclonal antibody targeting the CD20 antigen on B lymphocytes, leading to cell lysis through complement-dependent cytotoxicity and antibody-dependent cellular cytotoxicity. It is primarily used in the treatment of chronic lymphocytic leukemia and relapsing forms of multiple sclerosis[1]. Dacarbazine is an alkylating agent that acts as a DNA methylating agent, causing DNA damage and apoptosis in rapidly dividing cells. It is indicated for metastatic malignant melanoma, Hodgkin lymphoma, soft tissue sarcoma, and other cancers[2][5]. The combination has not been established as a standard regimen but may be considered investigational or off-label for certain hematologic malignancies where both drugs' mechanisms could provide synergistic antitumor effects.
