Overview

Ofatumumab is a fully human monoclonal antibody that targets the CD20 antigen on B-cells, leading to their depletion through antibody-dependent cellular cytolysis and complement-mediated lysis. It is approved for the treatment of chronic lymphocytic leukemia (CLL) and relapsing forms of multiple sclerosis (MS), including clinically isolated syndrome, relapsing-remitting disease, and active secondary progressive disease. Ofatumumab was originally developed by GlaxoSmithKline (GSK) and later acquired by Novartis for further development in autoimmune indications[1][2][4][5][6]. GSK2110183 is an ATP-competitive small molecule inhibitor targeting AKT1/2/3 kinases. It has been investigated in phase I/II clinical trials for various hematological malignancies, solid tumors, multiple myeloma, and in combination with other agents such as ofatumumab for CLL[8]. The combination "ofatumumab + GSK2110183" refers to investigational use where both drugs are administered together—typically studied in oncology settings such as CLL or other hematologic cancers.
