Ofatumumab + methylprednisolone is a combination regimen used primarily in the treatment of relapsed or refractory chronic lymphocytic leukemia (CLL). Ofatumumab is a fully human anti-CD20 monoclonal antibody that targets the CD20 molecule on B lymphocytes, leading to their depletion through complement-dependent cytotoxicity and antibody-dependent cellular cytotoxicity. Methylprednisolone is a synthetic glucocorticoid corticosteroid with potent anti-inflammatory and immunosuppressive properties; it works by mimicking endogenous glucocorticoids to suppress immune responses and inflammation. The combination has been studied in clinical trials for CLL patients who are relapsed or refractory to other treatments. Ofatumumab was developed by Genmab and GlaxoSmithKline (now GSK), while methylprednisolone is an established generic drug originally developed by Upjohn (now part of Pfizer). This regimen offers non-myelosuppressive therapy with high response rates in heavily pretreated CLL populations[3][8].
