Overview

Ofatumumab is a fully human monoclonal antibody that targets the CD20 antigen on B lymphocytes, leading to their depletion through mechanisms such as complement-dependent cytotoxicity and antibody-dependent cellular cytotoxicity. It is approved for the treatment of relapsing forms of multiple sclerosis (as Kesimpta) and was previously used for chronic lymphocytic leukemia (as Arzerra, now discontinued)[1][5][6]. SB-485232, also known as iboctadekin, is a recombinant human interleukin-18 (IL-18), an investigational immunostimulatory cytokine. It has shown antitumor activity in preclinical models and has been studied in early-phase clinical trials in combination with other agents for recurrent ovarian cancer[2][4]. The combination "ofatumumab + SB-485232" refers to the use of both drugs together; however, there are no published studies or approvals specifically for this combination. Each drug acts via distinct mechanisms—ofatumumab depletes B cells by targeting CD20, while SB-485232 stimulates immune responses via IL-18 signaling.
