Ofirnoflast is a synthetic small molecule that acts as a selective allosteric inhibitor of serine/threonine-protein kinase NEK7. By disrupting the interaction between NEK7 and NLRP3, it prevents the assembly and activation of the NLRP3 inflammasome, leading to reduced production of pro-inflammatory cytokines such as IL-1β and IL-18. This mechanism targets key inflammatory pathways implicated in diseases like myelodysplastic syndromes (MDS), post-procedural inflammation, pain, and other inflammatory conditions. Ofirnoflast is being developed by Halia Therapeutics for its anti-inflammatory potential, with ongoing clinical trials primarily in lower-risk MDS patients who are refractory to or intolerant of erythropoiesis-stimulating agents[1][3][4][7][9].
