Overview

OGG1 siRNA refers to small interfering RNA molecules designed to silence the expression of 8-oxoguanine DNA glycosylase 1 (OGG1). OGG1 is a bifunctional DNA glycosylase primarily recognized for its role in the base excision repair (BER) pathway, where it identifies and excises 8-oxoguanine (8-oxoG) lesions resulting from oxidative stress. Beyond DNA repair, OGG1 has been identified as a key mediator of inflammation; it recruits transcription factors such as STAT1 to the promoters of pro-inflammatory cytokines, including IL-1β, IL-6, and TNF-α. In research related to sickle cell disease (SCD), OGG1 expression is significantly elevated in monocytes and macrophages, contributing to the characteristic chronic inflammatory state. The use of OGG1 siRNA to knock down gene expression has been shown to block the induction of these cytokines in experimental models, suggesting that targeting the OGG1-STAT1 axis may offer a therapeutic strategy for managing inflammation in SCD and other oxidative stress-related conditions.
