Ogremorphin is a small molecule inhibitor of G-protein coupled receptor 68 (GPR68), also known as Ovarian Cancer G-protein-coupled Receptor 1 (OGR-1). Identified through a zebrafish developmental screen by researchers at the University of Maryland and Johns Hopkins, it is being developed as a potential treatment for glioblastoma multiforme (GBM). GPR68 is a proton-sensing receptor that is activated by the acidic extracellular microenvironment resulting from the Warburg effect in tumors. Ogremorphin blocks this survival pathway, inducing ATF4-dependent ferroptosis in GBM cells. Preclinical data suggests it is selective for cancer cells and acts synergistically with standard-of-care treatments like temozolomide and radiation.
