OGX-133 is a second-generation antisense oligonucleotide (ASO) designed to inhibit the expression of Insulin-like Growth Factor Binding Protein-5 (IGFBP-5). IGFBP-5 is a protein that plays a critical role in cell survival and is frequently overexpressed in various cancers, including prostate and breast cancer. In the context of prostate cancer, IGFBP-5 is associated with the progression to castrate-resistant prostate cancer (CRPC) and the development of resistance to hormone therapy and chemotherapy. By binding to IGFBP-5 mRNA, OGX-133 triggers its degradation, thereby reducing protein levels and potentially sensitizing tumor cells to apoptosis and standard-of-care treatments. The drug was part of a collaborative pipeline between Sonus Pharmaceuticals and OncoGenex Pharmaceuticals prior to and following their merger.
