OGX-225

OGX-225 is a second-generation antisense oligonucleotide (ASO) drug candidate that targets insulin-like growth factor binding protein 2 (IGFBP-2) and insulin-like growth factor binding protein 5 (IGFBP-5). The drug was developed by Ionis Pharmaceuticals and later licensed to OncoGenex Pharmaceuticals for oncology applications. OGX-225 works by downregulating IGFBP-2 and IGFBP-5 expression, which are proteins overexpressed in various cancers including prostate cancer and breast cancer and play roles in castration-resistant progression and treatment resistance. Preclinical studies demonstrated that OGX-225 inhibited cancer cell growth both in vitro and in vivo, induced apoptosis, and showed multi-targeting effects beyond just IGFBP-2 and IGFBP-5 inhibition. The drug was investigated primarily for breast cancer and prostate cancer but development was discontinued in the preclinical phase.
