**OH2 + HX008** is a combination of two investigational therapies for cancer: - **OH2**: a genetically engineered oncolytic herpes simplex virus type 2 (HSV-2). OH2 is designed to selectively replicate in tumor cells, leading to direct tumor lysis. It is further engineered to express human granulocyte-macrophage colony-stimulating factor (GM-CSF), which enhances antitumor immune responses by promoting immune cell infiltration and activation. - **HX008**: a humanized IgG4 monoclonal antibody targeting programmed cell death protein 1 (PD-1). It acts as an immune checkpoint inhibitor, blocking the interaction of PD-1 with its ligands PD-L1 and PD-L2, thereby reactivating tumor-infiltrating lymphocytes and enhancing immune-mediated tumor cell killing. In the combination, OH2 is administered by intratumoral injection, while HX008 is given systemically. The combination is under investigation for advanced solid tumors and melanoma, showing an acceptable safety profile and preliminary antitumor responses in early-phase clinical trials[1][2][3][4].
