**oHSV-IL12** is a genetically engineered oncolytic herpes simplex virus type 1 (HSV-1) designed to selectively infect, replicate within, and lyse cancer cells while simultaneously expressing the immune-stimulatory cytokine interleukin-12 (IL-12) within the tumor microenvironment[1][2][3][8]. Local IL-12 secretion enhances antitumor immunity by promoting Th1 differentiation, activating T cells and NK cells, increasing IFN-γ production, and turning the tumor environment from immunosuppressive to immunostimulatory. This multifaceted approach also inhibits tumor angiogenesis, further impairing tumor growth[1][2][8]. Multiple constructs exist (e.g., G47Δ-IL12, M002, NV1042, T-mfIL12), with G47Δ-IL12 among the most clinically advanced[1][6][8]. oHSV-IL12 is under development for difficult-to-treat cancers, including glioma, triple-negative breast cancer, and malignant peripheral nerve sheath tumors[1][2][8].
