oHSV-NIS is a recombinant oncolytic herpes simplex virus type 1 (oHSV-1) engineered to express the human sodium iodide symporter (NIS). It is designed for radiovirotherapy, a strategy that combines the direct cell-killing power of an oncolytic virus with the targeted delivery of radiotherapy. The virus selectively replicates in cancer cells, leading to oncolysis, while the expression of the NIS transgene allows infected cells to actively uptake and concentrate radioisotopes like Iodine-131. This dual mechanism facilitates non-invasive imaging of viral spread using SPECT/CT and enhances antitumor activity through localized radiation. Developed primarily at the Mayo Clinic, oHSV-NIS has shown significant potential in preclinical models of prostate cancer.
