Overview

oIL9R-engineered pmel-1 T cells are an experimental adoptive cell therapy consisting of T cells (typically CD8+ T cells from the pmel-1 transgenic mouse model) that have been genetically modified to express an orthogonal interleukin-9 receptor (oIL9R). This system utilizes a mutated IL-9 receptor that selectively binds to a corresponding synthetic (orthogonal) IL-9 ligand, which does not interact with the endogenous wild-type IL-9 receptor. This approach allows for the selective expansion and activation of the engineered T cells in vivo, bypassing the systemic toxicities associated with high-dose wild-type cytokine administration (such as IL-2). The pmel-1 TCR specifically recognizes the gp100 (PMEL) antigen, a lineage-specific protein expressed in melanocytes and melanoma cells. This technology was developed through a collaboration between the University of California, Los Angeles and Stanford University, and is being commercialized by Synthekine for the treatment of solid tumors.
