OJI-1 is a selective and potent small molecule inhibitor of histone deacetylase 8 (HDAC8). It is being investigated for the treatment of multiple myeloma (MM). HDAC8 is a class I HDAC isoenzyme that deacetylates lysine residues on histones and non-histone proteins like RAD51. By inhibiting HDAC8, OJI-1 increases RAD51 acetylation, which in turn inhibits homologous recombination (HR) activity and DNA break repair, leading to increased genomic instability and apoptosis in MM cells. Preclinical studies have shown that OJI-1 has a favorable therapeutic index and exhibits synergistic effects when combined with standard MM therapies such as dexamethasone, bortezomib, and lenalidomide.
