OKI-1546 is a small molecule inhibitor of Cyclin-dependent kinase 12 (CDK12) being developed by OnKure Therapeutics. CDK12 is a transcriptional regulator that plays a critical role in the expression of DNA damage response (DDR) genes. By inhibiting CDK12, OKI-1546 aims to induce a "BRCAness" phenotype in cancer cells, characterized by the downregulation of critical DDR proteins. This mechanism is intended to sensitize tumor cells to DNA-damaging agents, such as platinum-based chemotherapy or PARP inhibitors, or to induce synthetic lethality in specific genetic backgrounds. OKI-1546 is currently in preclinical development for the treatment of various oncology indications.
