**OKN-007 + temozolomide** is an investigational combination therapy for glioblastoma and recurrent high-grade glioma. OKN-007 is a nitrone-based small molecule (disodium 4-[(tert-butyl-imino) methyl] benzene-1,3-disulfonate N-oxide) with demonstrated anti-cancer activity, including reduction of cell proliferation and angiogenesis and increased apoptosis, while temozolomide is a DNA alkylating agent already approved for glioblastoma. Preclinical studies show OKN-007 enhances the effects of temozolomide, even in temozolomide-resistant cell lines, improving survival in animal glioma models. In clinical studies, the combination therapy is administered intravenously for OKN-007 and orally for temozolomide, and has shown manageable safety and promising improvement in progression-free and overall survival versus standard therapy.
