Overview

OKN4395 is a first-in-class, highly selective small molecule triple inhibitor targeting the prostanoid receptors EP2, EP4, and DP1. Developed initially by Idorsia Pharmaceuticals and now advanced by Owkin (and its affiliate Epkin), this investigational drug is designed to restore immune function in patients with advanced solid tumors. By blocking the immunosuppressive effects of prostaglandin E2 (PGE2) on EP2/EP4 receptors and prostaglandin D2 (PGD2) on DP1 receptors in immune cells, OKN4395 aims to enhance anti-tumor immunity. The drug leverages Owkin’s AI-powered K1.0 platform for asset selection and clinical development optimization. As of early 2025, it is being evaluated in Phase 1 clinical trials for various solid tumors including head and neck cancer, non-small cell lung cancer, squamous cell carcinoma, mesothelioma, sarcoma, colorectal cancer, dedifferentiated liposarcoma, and myxoid MFH.
