OKT3 + beta-D-glucosylceramide is an investigational oral combination therapy developed by the Hadassah Medical Organization. It consists of OKT3, a murine monoclonal antibody targeting the CD3 epsilon chain on T cells, and beta-D-glucosylceramide (GC), a naturally occurring glycolipid. The therapy is designed to induce immune tolerance by promoting the generation of regulatory T cells (Tregs) and modulating immune responses through the gut-associated lymphoid tissue (GALT). Unlike parenteral administration of OKT3, which is associated with severe cytokine release syndrome, the oral formulation is intended to be safe and biologically active. It has been investigated for the treatment of autoimmune diseases, nonalcoholic steatohepatitis (NASH), and metabolic conditions.
