OL-JC2 is a preclinical-stage small molecule agonist of Casein Kinase 1 alpha (CK1α) being developed by Oleolive for the treatment of pulmonary fibrosis, specifically idiopathic pulmonary fibrosis (IPF). The compound is designed to block TGFβ-driven fibrotic pathways locally in the lung. To address challenges with oral bioavailability and minimize systemic side effects, Oleolive is developing OL-JC2 as an inhalable therapeutic. The drug was discovered through a high-content imaging screen aimed at identifying compounds that could inhibit TGFβ activity, a central driver of fibrosis. The development has been supported by grant funding from the National Heart, Lung, and Blood Institute (NHLBI).
