Drug intelligence / Profile preview
TEV-749 is a long-acting injectable (LAI) formulation of the atypical antipsychotic olanzapine, currently in Phase 3 development by Teva Pharmaceuticals in collaboration with MedinCell. It utilizes the proprietary BEPO technology, a diblock and triblock copolymer-based drug delivery system that forms a biodegradable depot upon subcutaneous injection, allowing for sustained release of the active ingredient. The primary indication is the treatment of schizophrenia. TEV-749 is specifically designed to address the safety concerns associated with the currently marketed intramuscular olanzapine LAI (Zyprexa Relprevv), particularly the risk of Post-Injection Delirium/Sedation Syndrome (PDSS). By utilizing a subcutaneous route of administration and a different delivery matrix, TEV-749 aims to eliminate the need for the intensive post-injection monitoring period required by existing formulations. Olanzapine's therapeutic effect is mediated through a complex pharmacology involving antagonism at multiple receptor sites, most notably dopamine D2 and serotonin 5-HT2A receptors.
