Olaparib is a small molecule inhibitor of poly (ADP-ribose) polymerase (PARP), specifically targeting PARP1 and PARP2 enzymes involved in DNA repair. By inhibiting these enzymes, olaparib exploits the defective DNA repair mechanisms in cancer cells with BRCA mutations, leading to cell death. It is primarily indicated for the treatment and maintenance of BRCA-mutated ovarian, fallopian tube, peritoneal, breast, pancreatic, and prostate cancers. Olaparib was originally developed by KuDOS Pharmaceuticals and further developed by AstraZeneca following its acquisition; Merck & Co. has also been involved in its clinical development. The drug was first approved by regulatory agencies in 2014.
