By blocking PD-1 interaction with its ligands PD-L1 and PD-L2, pembrolizumab enhances immune-mediated tumor cell killing. The combination aims to exploit complementary mechanisms—DNA damage induction and impaired repair (olaparib), cytotoxicity via mitotic arrest (paclitaxel), and immune checkpoint inhibition to enhance antitumor immunity (pembrolizumab).
