Overview

Olaparib + bevacizumab is a combination therapy used as maintenance treatment for adults with advanced epithelial ovarian, fallopian tube, or primary peritoneal cancer whose tumors are homologous recombination deficiency (HRD)-positive, including those with BRCA mutations. Olaparib is a small molecule inhibitor of poly (ADP-ribose) polymerase (PARP) enzymes PARP1 and PARP2, which impairs DNA repair in cancer cells and induces cell death. Bevacizumab is a monoclonal antibody that inhibits vascular endothelial growth factor A (VEGF-A), blocking angiogenesis and reducing tumor blood supply. The combination leverages both direct cytotoxicity from impaired DNA repair and antiangiogenic effects to improve outcomes in HRD-positive cancers[1][2][3][4][5][6][8].
