Olaparib + pembrolizumab + bevacizumab is an investigational combination therapy comprising three agents with distinct mechanisms of action, used primarily in the maintenance treatment of ovarian cancer. Olaparib is a small molecule inhibitor of poly (ADP-ribose) polymerase (PARP), which impairs DNA repair in tumor cells with homologous recombination deficiency. Pembrolizumab is a monoclonal antibody that targets the programmed cell death protein 1 (PD-1) receptor, acting as an immune checkpoint inhibitor to enhance anti-tumor immune responses. Bevacizumab is a monoclonal antibody targeting vascular endothelial growth factor A (VEGF-A), inhibiting angiogenesis and thereby restricting tumor blood supply. This triplet regimen has been studied in BRCA wild-type and platinum-sensitive recurrent ovarian cancer, showing promising efficacy and manageable safety profiles[1][2][3][4][7].
