Overview

This entry refers to the combination of olaparib and placebo as used in clinical trials, typically as separate arms for comparison rather than a true pharmacological combination. Olaparib is a small molecule inhibitor of poly(ADP-ribose) polymerase (PARP), an enzyme involved in DNA repair. By inhibiting PARP, olaparib induces synthetic lethality in cancer cells with defects in homologous recombination repair pathways, such as those with BRCA1 or BRCA2 mutations. Placebo is an inert substance used as a control in clinical studies and has no therapeutic effect. In randomized controlled trials for cancers such as ovarian cancer, breast cancer, pancreatic cancer, and prostate cancer, patients may be assigned to receive either olaparib or placebo to assess the efficacy and safety of the active drug[1][10][7]. This "combination" does not represent a marketed co-formulation but rather reflects trial design.
