Overview
Olaratumab is a fully human IgG1 monoclonal antibody that selectively binds to the external domain of human platelet-derived growth factor receptor alpha (PDGFR-α) with high affinity, blocking ligand binding and inhibiting receptor activation. This action prevents downstream signaling involved in cell proliferation, chemotaxis, and mesenchymal stem cell differentiation. Olaratumab was developed for use in combination with doxorubicin to treat adults with advanced soft tissue sarcoma (STS) not amenable to curative treatment by surgery or radiation therapy. It received accelerated approval based on improved progression-free and overall survival but was later withdrawn from the market after confirmatory trials failed to show clinical benefit[1][3][5][6][7].
