Overview

This is a combination drug regimen consisting of **olaratumab** (a recombinant human IgG1 monoclonal antibody targeting platelet-derived growth factor receptor alpha, PDGFR-α), **gemcitabine** (a nucleoside analog and antimetabolite that inhibits DNA synthesis), and **docetaxel** (a taxane class chemotherapeutic that stabilizes microtubules and prevents cell division). This regimen has been investigated as a treatment for advanced soft tissue sarcomas (STS). The combination aims to build on the established efficacy of gemcitabine plus docetaxel in advanced STS by potentially adding targeted anti-tumor activity via olaratumab’s inhibition of PDGFR-α signaling. Olaratumab was developed by Eli Lilly. The ANNOUNCE 2 randomized phase 2 trial found that although safety was manageable and no new signals were observed, the addition of olaratumab did not significantly improve overall survival compared to gemcitabine plus docetaxel alone in this setting[1][3].
