Olaratumab + mitoxantrone + prednisone is a combination regimen studied for the treatment of metastatic castration-resistant prostate cancer (mCRPC) in patients who have progressed after docetaxel. - **Olaratumab** is a fully human monoclonal antibody that binds to **platelet-derived growth factor receptor-alpha (PDGFRα)**, blocking downstream signaling pathways involved in tumor growth and metastasis. - **Mitoxantrone** is an anthracenedione antineoplastic agent that intercalates into DNA and inhibits topoisomerase II, causing DNA strand breaks and inhibiting cancer cell replication. - **Prednisone** is a synthetic glucocorticoid used for its anti-inflammatory and immunosuppressive activities, and is commonly combined with chemotherapeutics in prostate cancer to help control symptoms and reduce some toxicities of chemotherapy. In a Phase II trial, this combination did not significantly improve progression-free survival compared to mitoxantrone and prednisone alone but showed an acceptable safety profile.
