olaratumab + nab-paclitaxel + gemcitabine

Olaratumab + nab-paclitaxel + gemcitabine is a combination regimen of three pharmaceuticals administered intravenously for the treatment of advanced malignancies, most notably evaluated in metastatic pancreatic ductal adenocarcinoma (PDAC). Olaratumab is a fully human monoclonal antibody targeting platelet-derived growth factor receptor alpha (PDGFRα), acting as an antagonist to block the receptor’s signaling that is implicated in tumor growth and metastasis. Nab-paclitaxel is a nanoparticle albumin-bound formulation of the antimitotic agent paclitaxel, which inhibits microtubule disassembly, while gemcitabine is a nucleoside analog that interferes with DNA synthesis, leading to cell death. This specific three-drug combination was evaluated in clinical trials in treatment-naïve patients with metastatic PDAC; although it demonstrated safety, it failed to significantly improve survival outcomes compared to nab-paclitaxel plus gemcitabine alone.
