Olaterepatide (PL9643) is a synthetic peptide melanocortin receptor agonist developed by Palatin Technologies for the treatment of dry eye disease (DED). It primarily targets the melanocortin-1 (MC1R) and melanocortin-5 (MC5R) receptors, which are expressed on the ocular surface and in the lacrimal glands. By activating these receptors, olaterepatide exerts anti-inflammatory effects and promotes the resolution of inflammation, potentially improving both the signs and symptoms of DED. Unlike many current treatments that focus on broad immunosuppression, olaterepatide leverages the body's natural anti-inflammatory pathways. It is currently being evaluated in Phase 3 clinical trials (e.g., MELODY-1) as an ophthalmic solution.
