Oleclumab is a human monoclonal antibody that targets the ectoenzyme CD73 (also known as 5'-nucleotidase or NT5E) on the surface of tumor and immune cells. By binding to CD73, oleclumab inhibits its enzymatic activity, which reduces the production of adenosine in the tumor microenvironment. Adenosine suppresses immune responses against tumors; thus, blocking CD73 with oleclumab can enhance antitumor immunity. Oleclumab is being developed primarily for cancer immunotherapy and has been studied in combination with other agents such as durvalumab. It was originally developed by MedImmune and AstraZeneca for use in various solid tumors including non-small cell lung cancer (NSCLC), pancreatic ductal adenocarcinoma (PDAC), breast cancer, sarcoma, and others[1][3][4][5].
