Overview

**Oligomycin** is a macrolide compound produced by Streptomyces species, primarily used as a research tool due to its potent inhibition of mitochondrial oxidative phosphorylation. Its mechanism of action centers on **blocking the proton channel (Fo subunit) of the mitochondrial F1Fo ATP synthase**, which halts ATP production by oxidative phosphorylation. This inhibition causes a substantial drop in mitochondrial ATP synthesis, impedes electron transport, and in turn induces processes like apoptosis and metabolic alteration in affected cells. Oligomycin is highly cytotoxic in mammalian cells but is also employed to probe mitochondrial function, cell metabolism, and as a toxin payload in experimental antibody-drug conjugates. It has no approved medical use due to toxicity[1][3][4][6][7].
