**Olintatug tesirine (ADCT-901)** is an antibody-drug conjugate (ADC) comprising a humanized monoclonal antibody (3A4) targeting **kidney-associated antigen 1 (KAAG1)**, a protein overexpressed in various solid tumors with restricted expression in healthy tissues, conjugated via a protease-cleavable linker to **tesirine (SG3249)**, a potent pyrrolobenzodiazepine (PBD) dimer cytotoxin. KAAG1 plays a key role in tumor cell proliferation, and the ADC internalizes upon binding, releasing the payload to form DNA interstrand crosslinks, inhibiting DNA replication and inducing tumor cell death. Initially developed by **ADC Therapeutics**, it targets advanced solid tumors with high unmet needs, including breast, ovarian, prostate, renal cell carcinoma, cholangiocarcinoma, and others.
