Olisutrigine bromide

ASN-008 (also known as Olisutrigine bromide) is a topical small molecule drug under development for the treatment of pruritus associated with atopic dermatitis and notalgia paresthetica. It belongs to the class of aminoindane compounds and acts as a sodium channel blocker, specifically targeting epithelial sodium channels (ENaC) on sensory/afferent neurons. ASN-008 is designed to selectively inhibit sensory nerve signaling involved in itch without affecting motor nerves, due to its entry into neurons via activated TRPV1 or TRPA1 channels that are predominantly expressed on sensory fibers. The drug is formulated as a gel for topical application and has demonstrated rapid onset and long duration of anti-pruritic action in preclinical models, with low systemic exposure observed in clinical studies. ASN-008 is currently in Phase 2 clinical trials for atopic dermatitis.
