Overview

**Olomorasib + carbamazepine** is a hypothetical combination of olomorasib, a potent, highly selective, second-generation small molecule inhibitor targeting the KRAS G12C mutation, and carbamazepine, an established anticonvulsant commonly used to treat epilepsy and neuropathic pain. Olomorasib irreversibly binds mutant KRAS G12C, inhibiting downstream oncogenic MAPK signaling in cancers such as non-small cell lung cancer (NSCLC). Carbamazepine acts as a voltage-gated sodium channel blocker, stabilizing neuronal membranes and reducing synaptic transmission. As of now, there is no reported clinical or preclinical development or usage of this specific combination, and no efficacy, safety, or rationale for pairing these drugs has been described in scientific literature or clinical trials[1][2][3][4][5].
