Olomorasib + cetuximab is an experimental combination therapy being investigated primarily for the treatment of advanced or metastatic KRAS G12C-mutant colorectal cancer (CRC). **Olomorasib** is a potent and highly selective, oral, second-generation KRAS G12C inhibitor that disrupts oncogenic KRAS-driven signaling. **Cetuximab** is a monoclonal antibody targeting the Epidermal Growth Factor Receptor (EGFR), thereby preventing EGFR-mediated signaling involved in tumor cell proliferation and survival. The combination aims to overcome compensatory feedback signaling through EGFR that can limit the efficacy of KRAS G12C inhibition alone, a mechanism particularly relevant in colorectal cancer. Early-phase clinical trials indicate that the combination has a higher objective response rate and improved disease control compared to single-agent KRAS G12C inhibition, with manageable toxicity.
