Overview

Olpasiran is a small interfering RNA (siRNA) therapeutic developed to lower lipoprotein(a) [Lp(a)] levels in patients at high risk for cardiovascular disease. It works by targeting and degrading apolipoprotein(a) messenger RNA (mRNA), thereby blocking the production of Lp(a), a major carrier of oxidized phospholipids implicated in inflammation and atherosclerosis. Clinical trials have shown that higher doses of olpasiran can reduce Lp(a) levels by more than 95% in patients with established atherosclerotic cardiovascular disease, with effects sustained for months after dosing. Olpasiran also leads to significant reductions in oxidized phospholipids on apolipoprotein B, which are believed to promote vascular inflammation and plaque formation. The drug is being developed by Amgen and is currently undergoing phase 3 clinical trials for the treatment of cardiovascular disease associated with elevated Lp(a)[1][3][5][6][7].
