Drug intelligence / Profile preview
OLT-201 is a topical gel formulation of dapansutrile (OLT1177), a selective small molecule inhibitor of the NLRP3 (nucleotide-binding oligomerization domain, leucine-rich repeat and pyrin domain-containing 3) inflammasome. Developed by Olatec Therapeutics, OLT-201 is designed to treat inflammatory skin conditions by blocking the activation of the NLRP3 inflammasome, which prevents the maturation and release of pro-inflammatory cytokines such as interleukin-1β (IL-1β) and interleukin-18 (IL-18). By targeting the innate immune system's inflammatory response at the source, OLT-201 aims to provide a localized anti-inflammatory effect with minimal systemic exposure. The drug has been primarily investigated in Phase 2 clinical trials for the treatment of mild to moderate atopic dermatitis, where it is evaluated for its ability to reduce skin lesions and associated symptoms like pruritus.
