Overview

**Olverembatinib + blinatumomab** is a combination regimen comprising a third-generation small molecule tyrosine kinase inhibitor (TKI), olverembatinib, and a bispecific T-cell engager antibody, blinatumomab. This regimen is primarily investigated for the treatment of Philadelphia chromosome–positive acute lymphoblastic leukemia (Ph+ ALL), ABL-class Ph-like ALL, and relapsed/refractory Ph+ ALL, including those with the T315I mutation. Olverembatinib inhibits BCR-ABL tyrosine kinase, effective against a broad spectrum of ABL1 mutations including T315I, while blinatumomab is a bispecific antibody that engages CD3-positive T cells to target and lyse CD19-positive B lymphoblasts. Clinical reports and small trials to date suggest the combination is highly effective, often producing high rates of complete and molecular remission with manageable toxicity, and it may offer a chemotherapy-free treatment strategy for these leukemias[1][2][3][5].
