Drug intelligence / Profile preview
Olverembatinib + lisaftoclax is an investigational, oral, chemotherapy-free combination regimen developed by Ascentage Pharma. Olverembatinib is a potent third-generation tyrosine kinase inhibitor (TKI) that targets BCR-ABL1 (including the T315I mutant and other resistant variants) and KIT. Lisaftoclax is a novel, selective small-molecule BCL-2 inhibitor. Together, they act synergistically to increase apoptosis and reduce anti-apoptotic proteins such as BCL-2, BCL-xL, and MCL-1. The combination is primarily being evaluated in clinical trials for relapsed/refractory Philadelphia chromosome-positive acute lymphoblastic leukemia (Ph+ ALL), including pediatric populations, and advanced chronic myeloid leukemia (CML) or Ph+ acute myeloid leukemia (AML).
