Drug intelligence / Profile preview
HQP1351 + APG-2575 + homoharringtonine is an investigational combination regimen consisting of three active agents: olverembatinib (HQP1351), a third-generation BCR-ABL tyrosine kinase inhibitor that also targets FLT3 and KIT; lisaftoclax (APG-2575), a selective BCL-2 inhibitor; and homoharringtonine (HHT), a plant-derived protein synthesis inhibitor. Developed by Ascentage Pharma, this triplet regimen is being evaluated in Phase Ib clinical trials for the treatment of patients with relapsed or refractory acute myeloid leukemia (R/R AML), particularly those who have failed prior treatment with venetoclax or other BCL-2 inhibitors. The therapeutic rationale relies on the synergistic induction of apoptosis; homoharringtonine-mediated protein synthesis inhibition downregulates MCL-1, a key mediator of resistance to BCL-2 inhibitors, while olverembatinib's FLT3 inhibition further suppresses MCL-1, thereby enhancing the pro-apoptotic activity of lisaftoclax.
