Olverembatinib + venetoclax + dexamethasone (OVD) is an investigational, chemotherapy-free combination regimen being evaluated for the treatment of Philadelphia chromosome-positive acute lymphoblastic leukemia (Ph+ ALL). The regimen combines olverembatinib, a third-generation tyrosine kinase inhibitor (TKI) that targets BCR-ABL1 (including the gatekeeper T315I mutation), with venetoclax, a BCL-2 inhibitor that promotes apoptosis, and dexamethasone, a corticosteroid. This triplet therapy aims to provide a potent, targeted approach to Ph+ ALL, potentially reducing the toxicity associated with traditional intensive chemotherapy. Clinical studies, particularly in newly diagnosed patients, have shown high rates of complete remission and minimal residual disease (MRD) negativity with manageable hematologic toxicities.
