OLX75016 is an investigational small interfering RNA (siRNA) therapeutic developed by OliX Pharmaceuticals for the treatment of metabolic dysfunction-associated steatohepatitis (MASH), also known as nonalcoholic steatohepatitis (NASH), with accompanying liver fibrosis and for weight management. The drug utilizes proprietary RNA interference (RNAi) technology, specifically GalNAc-conjugated asymmetric siRNA (asiRNA), to down-regulate the expression of disease-causing genes implicated in liver fat accumulation, inflammation, and fibrosis. Preclinical studies have shown that OLX75016 reduces liver fat content, inflammation, and fibrosis in obese models. It has demonstrated enhanced weight loss effects when combined with GLP-1 receptor agonists such as semaglutide and may mitigate weight rebound after cessation of GLP-1 therapy. The mechanism is differentiated from existing therapies by targeting gene expression rather than acting directly on metabolic receptors or hormones[1][5][6][8].
