Drug intelligence / Profile preview
OM-302 is a preclinical-stage synthetic fusion peptide being developed by Oncolyze as a back-up candidate to OM-301 (PNC-27) for the treatment of various cancers, including acute myeloid leukemia (AML), melanoma, pancreatic, breast, colon, ovarian, and lung cancers. Derived from the HDM2-binding domain of the p53 tumor suppressor protein (specifically residues 17-26) fused to a membrane-penetrating peptide (membrane residency peptide or MRP), OM-302 targets cell-surface HDM2 (human double minute 2) on cancer cells. Upon binding to membrane-bound HDM2, the peptide undergoes a conformational change that induces transmembrane pore formation (poration), leading to rapid cell lysis and necrosis (a process termed 'pop-tosis' or necrobiosis) in a p53-independent manner, while sparing normal cells.
