Overview

OM102 is a multi-armed oncolytic myxoma virus (MYXV) candidate developed by OncoMyx Therapeutics for the treatment of advanced solid tumors. Myxoma virus is a leporipoxvirus that is naturally tumor-selective due to its reliance on pathways commonly activated in cancer cells, while remaining non-pathogenic to humans. OM102 is engineered to carry three distinct immunomodulatory payloads: interleukin-12 (IL-12) to promote Th1 immune responses and activate T and NK cells; decorin, a proteoglycan that inhibits TGF-beta and modulates the extracellular matrix; and a CD47 blocker (SIRPα-Fc) to inhibit the "don't eat me" signal and enhance macrophage-mediated phagocytosis. This combination is designed to transform the immunosuppressive tumor microenvironment into an immunostimulatory one, facilitating both local oncolysis and a systemic anti-tumor response.
