Ombrabulin + bevacizumab is an investigational intravenous combination therapy evaluated for the treatment of advanced solid tumors. **Ombrabulin** is a vascular disrupting agent (VDA) and a tubulin polymerization inhibitor that causes selective shutdown of tumor vasculature by disrupting endothelial cells, leading to tumor necrosis. **Bevacizumab** is a monoclonal antibody that inhibits vascular endothelial growth factor A (VEGF-A), thereby preventing new blood vessel formation (angiogenesis) in tumors. The rationale for combining these drugs is based on their complementary mechanisms: ombrabulin rapidly collapses existing tumor blood vessels, while bevacizumab blocks the formation of new vasculature. Clinical studies (Phase I) sought to establish the safety, pharmacokinetics, and recommended dosing for the combination. The development of ombrabulin was discontinued after lack of efficacy in later trials, thus this combination is not marketed.[1][3]
