Ombrabulin + cisplatin is a **combination therapy** consisting of the vascular disrupting agent **ombrabulin** (AVE8062, an analog of combretastatin A4) and the platinum-based chemotherapeutic agent **cisplatin** (CDDP). Ombrabulin acts by disrupting established tumor blood vessels, resulting in rapid shutdown of tumor perfusion and necrosis. However, because a rim of viable tumor cells often persists when ombrabulin is used alone, it is combined with cytotoxic agents like cisplatin to enhance antitumor efficacy. Cisplatin functions as a DNA-alkylating agent, forming DNA cross-links and inhibiting DNA replication and transcription, leading to apoptosis of cancer cells. This combination is studied primarily for the treatment of advanced solid tumors and has demonstrated manageable toxicity profiles and preliminary antitumor activity in phase I clinical trials, though some studies terminated early due to toxicity in specific populations.
