Overview

Ombrabulin + paclitaxel + carboplatin is an investigational combination therapy consisting of the vascular disrupting agent (VDA) ombrabulin and the standard chemotherapy doublet paclitaxel and carboplatin. Ombrabulin (AVE8062) is a water-soluble analog of combretastatin A4 that binds to the colchicine site of tubulin, inhibiting polymerization and leading to the destruction of tumor vasculature and subsequent tumor necrosis. Paclitaxel is a taxane that stabilizes microtubules, while carboplatin is a platinum-based alkylating agent that causes DNA cross-linking. Developed by Sanofi, this combination was evaluated in Phase II and III trials for advanced solid tumors, including ovarian, cervical, and non-small cell lung cancers, but development was discontinued due to a lack of significant clinical benefit.
